New nano-transistors directly detect cell interiors

According to the American Physicist Network and the British "Nature" magazine website reported on August 12, Harvard University chemists and engineers jointly produced a new V-shaped nano-transistor with a layer of phospholipid bilayer. It is very easy to enter the inside of the cell for detection without causing any visible damage to the cells.
The new device, called nanoscale field-effect sensors or nano-FETs, is also described in detail in this week's issue of Science. It is smaller than many viruses. Human cells range in diameter from about 10 micrometers to 50 micron heart cells, while new sensors measure cells in the 5 micron diameter. There are several series of nano FETs, the overall size is less than 50 nanometers, and the nanowire needle diameter is only 15 nanometers.
The current glass pipettes used to record electrical signals in nerve cells and other cells are only micron-sized with an electrode attached to the cell membrane to record electrical signals. But the technology is far from ideal, and Charles Leb, a chemist at Cambridge University in Massachusetts and Harvard University who led the study, said the pipettes are cumbersome and often damage cells when they work.
The surface of the nano-FETs is coated with a phospholipid bilayer membrane, which is the same as the composition of the cell membrane. It can penetrate into the cell without any effort by cell membrane fusion, which is similar to the process of phagocytosis of viruses or bacteria. This means that the insertion of nano-FETs leaves almost no wounds, it can enter and stay in the cells and be continuously detected.
Nano-FETs have a three-dimensional flexible structure. Two coated nanowires are fixed at the top of the V-shaped 60-degree angle, and are connected at an angle of 120 degrees to the main body. When working, there is current passing through the nano-transistor. It can be used to detect ion changes and electronic signals in cells, especially nerve cells; it can also be immobilized on receptors or ligands to detect biochemical processes inside single cells. The team demonstrated the use of nano-FETs, which used a single cultured embryonic chicken heart cell to record a series of voltage peaks at a frequency of 2.3 Hz, consistent with cell beat rhythms.
The researchers said that this is the first method of testing semiconductor devices inside a cell in 10 years, and it is a new method of intracellular research. His team has used the device for nerve cells and is working with the MIT research team to study the growth of tissues linked to nanowires.
LeBow said that their long-term goal is to combine digital and biological research to produce artificial tissue and implant nanowires to enhance medical monitoring.
